Alternative communication system during disaster Essay

PSTN or the public switched telephone network is the prime network responsible for transmitting and receiving voice, fax and data. All government and private offices, police stations, fire stations, hospitals and majority of homes and business places are serviced by the PSTN line. . Amateur Radio operation does not use ground based infrastructure. Moreover these have limited power needs which can easily be met by batteries and generations. Therefore, it has been experienced that Amateur Radio has worked successfully when no other communication worked. A radio wave is an electromagnetic wave propagated by an antenna. The floods in 2004 in Assam and Bihar submerged telephone exchanges and damaged telephone exchanges thereby disrupting roads and railways communication. This is the reason why the district headquarters got totally out off from the state headquarters and neighbouring districts. A major natural disaster or emergency situation like earthquake, cyclone, flood, etc., leaves the public wired and wireless mobile telephone infrastructure damaged and non-functional. This is a transmission towers or disrupted power supply to operate telephone exchanges and cellular transmission towers also affect the radio communication network of police and civil authorities. Satellite based communication systems refer to communication systems designed for users on the earth but have some equipment in space called a satellite. Various jobs such as taking weather pictures or finding accurate positions are amongst the different job performed by different satellites. These are sometimes refered to as COSMAT’s and are essentially radio relay stations in space. Geo-stationary orbit satellites are stationed above the earth at a position around 36000 kms above the equator. The images of weather forecasts and cyclone observations that we see on our television are made with the help of images from weather satellites. A major disaster (flood, earthquake, etc.) or emergency situation affects telephone exchanges and communication networks thereby disconnecting the affected areas from other parts of the world. In such situations, the urgent needs of disaster affected population are not communicated to government authorities, voluntary organizations and other unaffected people. This result in severe loss of human lives, livestock, property etc. This situation calls for reliable alternate communication links, to ensure that the right resources are provided to the right people at the right time. Asynchronous Orbit Satellites – These satellites are positioned in the lowerorbits around the earth. These continuously change their position with respect to the earth. The different kinds of asynchronous orbit satellites are : a) Science satellites: These orbit in altitudes of 4800 to 9700 kms. These send their research data to earth via radio telemetry signals. b) Observation Satellites: These are used for tasks like search and rescue satellites, photography. These act as relay stations to rebroadcast emergency radio-beacon signals from a downed aircraft or ship in trouble. These orbit at altitudes from 480 to 970 kms. c) Global Positioning System Satellites: Orbiting at altitudes of 9600 to 19200 kms, these are used to determine in terms of latitude ad longitude to determine the exact geographic location. The two prime communication networks of government of India are NIC and BSNL. Following are the functions of them: NIC or the National Information Centre, an organization of the Government of India is a premier science and technology organization of the Government of India is a premier science and technology organization in the field of informat ics services and information technology applications. To facilitate information and communication applications in government departments at central, state and districts, it has established a nationwide ICT network i.e. NICNET. The Government has designated NICNET as the Government Network. It has gateway modes in Central Government Departments, 35 state and union territories secretariats and 602 district collect orates of India for IT services. BSNL : The Department of Telephone Operations, Government of India was incorporated on 1st October, 2000 and was named Bharat Sanchar Nigam Limited. If only the social and economic development is taking place, that is referred to as â€Å"equitable†, whereas, if only the environmental and economic development takes places, that is referred to as â€Å"viable† (Chick and Micklethwaite 83). CSR is achieved only when all three dimensions d evelop simultaneously; that is, it is bearable, equitable and viable (Chick and Micklethwaite 83). The very mention of CSR calls into question the reason for business’ existence. Whether or not businesses should be responsible for the society and environment remains questionable since the primary purpose of a business is to satisfy its shareholders and generate profits. This is the line of argument used by classical theorists who are against the practice of CSR. According to Milton Friedman, the only social responsibility of a corporate entity is that of doing business in an open competitive marketplace that is free of deception and any fraud by utilizing its resources in order to increase profits (Friedman 6). The classical advocates further maintain that managers are mainly responsible for catering to shareholders’ interests by enhancing financial returns to the business. Practicing CSR may lead to the dilution of economic productivity since the investments made in C SR may be irrecoverable (due to the non-profit nature of CSR activities) (Friedman 6). However, advocates of CSR base their arguments in favor of the same for various reasons. Usually, companies engage in such disclosure for two main reasons: achieving a competitive edge by enhancing employee morale and goodwill and pressure by external stakeholders such as governments, environmental agencies etc. Not adhering to the latter’s demands would mean a loss in company’s goodwill and, hence, profits. This leads us to the argument related to stakeholder view used by proponents of CSR. This view was conceptualized as early as in the 1970’s by Freeman who described organizations in terms of the managers’ relationships with stakeholders (Garriga and Mele 59). A stakeholder is typically any individual entity or group that has an impact on, or is affected by the corporation’s goals and objectives (Garriga and Mele 59). For instance, man can decide not to consume any corn by avoiding planting it. At the same time, corn cannot decide on whom to have it planted. The marriage between man and corn can be viewed from the perspective of man having first domesticated the wild plant. This was later followed by selection and breeding of the best variety, were it not for man, it would be wild up to now (Pollan, 2007). In Africa and some parts of Asia, the grits from maize are ingested by humans. For instance in central and eastern Africa, the grits are used to make a staple food called ugali. This food is easily made and readily consumed with wide range vegetables. Grits can also be used to make porridge that is highly consumed by young children especially in Africa and some parts of Asia. This food product is believed to provide energy to the children who have higher energy requirements in general. Corn can also be used to make bi fuel. This is presently being done in the US of and is hastily replacing the fuels that are pollutants of the atmosphere. Corn is preferred to refined oil because of its availability and environmentally sound aspects. It is also used to make the brewers grits that are used in the beer industry. This is because it contains starches and sugars that undergo fermentation when provided with the required conditions. The medicine industr y also makes utilize corn in their processed form. How is it possible that she can be seen in such contrasting perspectives? E.T. Donaldson proposes a solution that Chaucer â€Å"discloses a world in which humanity is prevented by its own myopia, the myopia of the describer, from seeing what the dazzlingly attractive externals of life really represent† (935). However, if the Wife is everyman and everywoman, then all of these perspectives can be true. Rather than a singular, marginalized character with limiting aspects, the Wife is a complex and comprehensive blend of Chaucer’s creations, and the center to which all of the other pilgrims and their tales return. Her portrait is more descriptive than any other character portrayed, but it also demonstrates the characteristics uniquely identified with each of the other characters. The prologue to her tale is the longest of any of the other characters. Is she just long-winded and full of prideful arrogance? I suggest that the limited details each of the other characters possess emanates from the comprehensive detail of the Wife’s portrait and prologue which completes a circular exposà ©. I propose that Chaucer’s Wife is not just a Wife, or even a woman; she is an amalgamation of literary possibilities. Her literary knowledge is suspect, but her worldly experience spans a lifetime through which she acquires autonomy. In the introduction to The Wordsworth Chaucer, Editor Larry Benson discusses how Chaucer the poet draws on his own literary experience in creating his tales, and so too does Allisoun of Bath possess the experience that allows her to transcend the limits of definition (11). She is deaf in both ears and therefore cannot hear the hyperbole of dominant male discourse. She does not argue her right to speak, she just speaks. Contrary to acceptable femininity, she is a hypersexual woman, but sex is neither for enjoyment, nor procreation. Finally, Chaucer is not writing as a woman, or as a man attempting to write as a woman or using masculine writing to portray a female, he is writing in a manner that is useful to educate everyone to limitless possibilities. The Wife of Bath then, rather than a singly signified representation based on scholarship perspective, is, in reality, the nexus of the Canterbury Tales. Although like the Wife there are limitless possibilities for proving this argument, it is my plan to defend this thesis through the tropes of status, voice and experience as seen in her portrait, and the prologue to her tale. To prepare the way for the convergence of his themes, where everything and everyone emanate from and return to a central focus in the Wife of Bath, a General Prologue begins the tales with a representation of the biblical creation story in its hierarchical progression. In the opening lines, first there is water and wind, plants, followed by animals, and finally, portraits of the travelers are depicted. It is spring, a time of new beginnings, as well as a time of pilgrimage to give thanks for past favors, and in Chaucer’s fourteenth century world, twenty-nine diverse pilgrims â€Å"with ful devout courage† (22), gather at a public inn to sojourn to Canterbury, site of the holy shrine of the martyred Thomas à   Beckett. Born in Cheapside, London, Be cket’s inclusion in the tales illustrates human diversity, from vacuous impoverishment to reverential saintliness, themes also recurring throughout the tales and demonstrated by the Wife’s portrait and prologue. Twenty-four pilgrims are presented in the portraits which begins with the highest rank, â€Å"a Knyght ther was, and that a worthy man† (43), and ends with the lowliest, â€Å"a gentil Pardoner† (669). According to a popular websites, Alisoun, a name which means â€Å"of nobility†, and â€Å"sacred flame† (1), is also a common Middle English name. The name given by Chaucer to the Wife, it is apparent from the multiplicity of meanings that Alisoun represents both high and low culture, a correlation to the cross-cultural representations of her fellow travelers. Taken together, the pilgrims represent every facet of society, facets seen inclusively as the Wife’s total composition. Each line of her portrait represents an attribute or characteristic illustrated in the portrait of one of the other pilgrims. While some of the travelers mimic one another in style or attribute, no other pilgrim possesses all of the qualities signified in each of the others. That Chaucer the poet repeated his themes over and over is not surprising, but that those themes all converge in the Wife of Bath is significant since it centers privilege in a woman. If the Wife is the Nexus, then Chaucer’s allusion to the creation story points to her as the Eve from whom all women are descended. While Eve is not mentioned in the prologue to the Canterbury tales, she is a pivotal component of the creation story to which Chaucer alludes. Chaucer would have wished to avoid accusations of heresy, a realistic concern in his time period. To have openly equated Eve with the Wife, with specificity of declaration, would have opened him up to, at the very least, severe criticism, which was not what he was interested in. He was interested in opening up the minds of the people by writing alternative realities. However, there are two prominent women in Chaucer’s prologue, why wouldn’t the Prioress be Eve? The Prioress is dainty, prepossessing and high in the social strata. Her portrait appears before the Wife’s portrait in the general prologue. She has beautiful manners and is very sympathetic to even the smallest creature. Additionally, the Prioress is a nun, a woman who swears obedience to authority when she takes her vows. She is not a good prototype for Eve since the Prioress is certainly willing to acquiesce, whereas Eve and the Wife are not. The Wife is strong, bold, does not care what people think of her, she is her own person and like Eve, who was unwilling to accept the dictates of authority and wanted to control her own destiny, the Wife is also in search of control over her life. While Eve is not specifically referenced in Chaucer’s creationism allusion, she is definitely alluded to by virtue of the hierarchical progression of pilgrims that Chaucer introduces to his readers. Adam and Eve occur in natural hierarchical progression in the creation story, from the lowest to the highest, water to man/woman, similar to the Chaucerian hierarchy in which the Wife is introduced to the reader. A compelling testament to the argument of the centrality of her character as binder for the tales is witnessed in the make-up of this very Chaucer class, where approximately sixty percent of the students have chosen to write their term paper on some characteristic aspect of the Wife. No two representations are even similar, and analysis of her character ranges from psychological to pornographical arguments with everything in-between. The very diversity of these contemporary arguments do, however, represent the critical diversity argued over the past seven centuries but existing scholarship fails to recognize her important thematic centrality.

In Ians note, structural theory suggest we are influenced in our behaviour and actions by society and adopt a macro approach. The macro level have an impact on the way Albi is treated by classmates or even when he is out for dinner with his family. Staff doesnt put in the effort to communicate with Albi and tends to ignore him which makes him feel like he does not exist. This have led to the conflict theory. Parents teaches their childrens of the norms however they can pick up bad habits while they are in school. He is being bullied because of his condition and he is isolated from other childrens as they see this as unn atural and abnormal. The troubling of it is that Albi is constantly in a fight which his parents are very worried. Due to the conflict between Albi and his classmates he feels unwanted and would rather stay at home. He has distanced himself from his friends which caused him not having any friends anymore. Another structural theory that is link to Albis change of behaviour is the symbolic theory. Childrens misuse of words such as deaf can you hear me or you cant hear, you must be stupid affects Albi emotionally, which he feels down and worthless. In Albis case this is very hard for him to cope as he is only a child, he wouldnt be able to understand the situation and his actions is uncontrollable. This can be difficult for his parents to manage as he might become disobedient. This can have an effect on him when he grows up, he will repeat these phrases to himself and can have a negative result later on his life. Once Albi starts attending the community centre for the deaf his private troubles will eventually improve. Everything will be new his life style will change and he will have to adjust, it may take a while for him to progress however he will get there in the end with the right kind of support from his loved ones. As human beings we strives to seek for new motivation therefor this will help him. Albi should be able to cope and learn quickly as his parents will be there to support him all the way through.ÂÂ   He will meet new people who has the same condition as him which will have a positive effect on him emotionally and make him realise that he is not alone in this battle. This will also help him to realise that being different from other childrens is ok. He will feel sense of belonging and understanding of his situation. He will feel wanted as he is in the same group of people and also because he will be part of the group activities or playing with someone without getting judge d. He will be trying to improve himself to fit back in with society with the support of staff, family and his new friends. This can improve the way he thinks of the world and his future. He is more likely to proceed in education which will have a good impact later on in his life. Albi is more likely to succeed in life and will be determined to success. The possibility of Albi not letting his condition get in the way of success is very high with the help from people who is important to him and the acceptance from others that he will receive as he grows older. To improve Albis life chances, the media could increase the advertisement about deafness. Making it clear to everyone that their situation is difficult and by saying or doing negative stuff towards them wont make things better. Perhaps making up a cartoon series about someone being deaf and treating them in the right manners could help children understand better. Making them feel that they belong with society can prevent horrible things that they can possible do to themselves. The schools can also help by teaching childrens that not everyone is born perfect and that they should still accept them for who they are. Schools can do group activities where they will get to experience how difficult it is not being able to hear so they will be able to understand and will unlikely treat their classmates in a disrespectful manner. Teachers can help by teaching the students to treat others the way they would like to be treated. In college we can create a power point all about deafness and how i t is important to respect them and that they should be treated the same. I could then present my presentation to different schools so they can have a better understanding of what deaf people have to go through each day as this can have a bad impact on themselves. For some they would like to end their life as people feeds them with negativity which does not help as they are already struggling and we are making no effort on how we could help rather us focus on how we can make them feel horrible about their self instead.

Assembly of Functional Cellulolytic Enzymes In the present study, we reported the assembly of functional cellulolytic enzymes using a synthetic, cell-surface engineered yeast consortium. Trichoderma reesei endoglucanase II (EGII) and cellobiohydrolase II (CBHII) and Aspergillus aculeatus ÃŽ ²-glucosidase I (BGLI) were displayed as fusion proteins with the AGA2p C-terminus of a-agglutinin on the cell surface of the diploid yeast strain Saccharomyces. cerevisiae Y5. The immobilization of each enzyme on the cell surface was confirmed by immunofluorescence microscopy. This type of yeast consortium allowed convenient optimization of ethanol production by adjusting the combination ratios of each cell type for inducing synergy in cellulose hydrolysis. Next, the direct ethanol fermentation from steam-exploded corn stover was investigated. The optimized cellulase-displaying consortium produced 20.4 g/l ethanol from 48.4 g cellulose per liter after 72 h in the presence of a small amount of cellulase reagent (0.9 FPU/ml). These findings suggested the feasibility of the cellulase-displaying yeast consortium for simultaneous saccharification and fermentation. Currently, many technological barriers exist with respect to the economical production of ethanol from lignocellulosic biomasses [1]. In the process of hydrolyzing cellulose into soluble sugars, multiple cellulases including endoglucanase (EG), cellobiohydrolase (CBH), and ÃŽ ²-glucosidase (BGL) are required [2]. Consolidated bioprocessing (CBP), which combines enzyme production, hydrolysis, and fermentation in one step, is a promising strategy for effective ethanol production from lignocellulosic materials. Saccharomyces cerevisiae is the traditional microorganism used for ethanol production, but it is unable to utilize cellulosic materials and a saccharification process is required prior to fermentation to produce glucose [3-4]. Numerous attempts have been made to engineer S. cerevisiae strains to express cellulases by cell surface engineering for direct ethanol production from cellulose, and although various bifunctional or trifunctional cellulose-degrading strains have been const ructed, the efficiency of cellulose degradation has not been sufficiently improved [5-9]. It would appear that co-expression of all cellulolytic enzymes in a single cell resulted in relatively low expression levels of cellulases, which may have been due to the heavy metabolic burden and potential jamming of the secretion machinery [6,7,10]. Therefore, in this study, we adapted a new strategy of performing simultaneous saccharification and fermentation with a synthetically engineered yeast consortium having the desired properties of cellulolytic ability and ethanol production to reduce the metabolic burden. The development of a diploid yeast strain is another promising strategy for improving expression levels of heterologous genes and enhancing the fermentation performance of S. cerevisiae. Because diploid strains have better growth ability as well as stress tolerances compared with haploid strains, they are particularly suited for industrial applications. Previously, our group reported on the construction of an à Ã‚ °-agglutinin expression system for genetic immobilization ÃŽ ²-glucosidase I on the cell surface of S. cerevisiae Y5 (Patent No: ZL200810222897.7, CGMCC2660). This diploid robust yeast strain possessed many advantages, such as higher ethanol yield, higher resistance to ethanol, and higher physiological tolerance to inhibitors present in lignocellulosic hydrolysates. Here, we report on our efforts to demonstrate the assembly of functional cellulolytic enzymes using a synthetic yeast consortium. In this study, we demonstrated the feasibility of constructing a novel cell surface engineered diploid yeast consortium for direct ethanol production from phosphoric acid swollen cellulose (PASC) and steam-exploded corn stover (CS), an important step toward direct ethanol production from insoluble cellulosic materials. The strains and plasmids used in this study are summarized in Table S1. Saccharomyces cerevisiae Y5 used for the yeast cell surface display of the cellulolytic enzymes was a newly developed diploid strain in our laboratory. E. coli Top 10 was used as the host strain for recombinant DNA manipulation. T. reesei was purchased from CICC (China Center of Industrial Culture Collection). E. coli transformants were grown in Luria-Bertani medium (1% tryptone, 0.5% yeast extract and 1% NaCl, pH 7.0) supplemented with 100 ug/ml of ampicillin. S. cerevisiae Y5 transformants were selected and maintained on Geneticin plates (1% yeast extract, 2% peptone and 2% glucose supplemented with 600 ug/ml Geneticin) at 30 °C , were induced in YPG (1% yeast extract, 2% peptone, and 2% galactose) at 20 °C. The fermentation medium was composed of 10 g/l yeast extract, 20 g/l polypeptone and 10 g/l PASC as the sole carbon source. The à ¯Ã‚ ¬Ã‚ lamentous fungus T. reesei was cultured in potato dextrose aga r medium (2% potato extract, 2% glucose) at 27 °C. The cDNA was synthesized from mRNA by using the First-Strand cDNA synthesis kit (Fermentas). Unless otherwise indicated, all chemicals, media components and supplements were of analytical grade standard and obtained from Sigma-Aldrich (St. Louis, MO, USA). All restriction enzymes were purchased from New England BioLabs (Ltd. Beijing). Primers used for plasmid construction are provided in Table S2. Plasmid pAGA1 for over-expression of the AGA1 gene and plasmid pBGLI for cell surface display BGLI were constructed previously [11]. Plasmid pEGII for cell surface expression of the EGII (egl2) was constructed as follows. The 1194 bp DNA fragment encoding the egl2 gene without its native secretion signal was amplià ¯Ã‚ ¬Ã‚ ed with the à ¯Ã‚ ¬Ã‚ rst-strand cDNA prepared from T. reesei as the template using primer pairs egl2-For/Rev, this DNA fragment was introduced into the yeast display vector pYD1(Invitrogen) with Kpn I/BamH I. MAT terminator was amplified from pYD1 by using primer pairs MAT-For/Rev and then digested with BamH I/EcoR I to create plasmid pYD1-egl2MAT. The KanR fragment was obtained from plasmid YIP5-KanR by two-step cloning. First, the DNA fragment containing ADH promoter and KanR ORF was amplified from YIP5-KanR by PCR using the KanR-For/Rev primers and inserted into EcoR I/Apa I site of plasmid pYD1-egl2MAT; next, the ADH terminator digested with Bgl II/Nde I was also introduced into pYD1-egl2MAT. The resulting plasmid was named pEGII. For displaying the T. reesei CBHII gene (cbh2) in S. cer evisiae Y5, plasmid pCBHII was created. A 1344 bp gene fragment coding for the mature region of the CBHII was amplià ¯Ã‚ ¬Ã‚ ed using primers cbh2-For/Rev-KT and introduced into plasmid pEGII digested with Kpn I/BamH I for replacing egl2 to form pCBHII (Figure 1). Transformation of S. cerevisiae Y5 was carried out using the lithium acetate method [12]. The plasmid pAGA1 was linearized by Apa I for chromosome integration. The plasmid pYD1 was transformed into S. cerevisiae Y5 as a negative control. S. cerevisiae Y5 clones transformed with different plasmids (strain Y5/pYD1 contained plasmids pAGA1 and pYD1, strain Y5/EGII contained plasmids pAGA1 and pEGII, strain Y5/CBHII contained plasmids pAGA1 and pCBHII) were selected and maintained on Geneticin(G418) plates. Immunofluorescence microscopy was performed as described previously [13]. Immunostaining was performed as follows. Induced recombinant yeast cells expressing cellulases were harvested by centrifugation at 6000 rpm for 5 min and washed with phosphate-buffered saline (PBS). As the primary antibody, mouse anti-Xpress tag antibody (Invitrogen, R910-25) for EGII and CBHII was used at dilution rates of 1:1000. As the second antibody, Fuorescein (FITC)-conjugated goat anti-mouse IgG(H+L) (Jackson, 115-095-003) was used at dilution rate 1:200. Cells and the anti-body were incubated at room temperature. After washing the cell–antibody complex with PBS twice, cellular localizations of the cellulases were observed under a fluorescence microscope. Yeast strains Y5 and Y5/pYD1were used as control. Yeast cells were induced in YPG medium for 48 h at 20 ºC and harvested by centrifugation for 5 min at 6000 rpm, washed with distilled water. BGLI activity of strain Y5/BGLI was measured using à ¯Ã‚ Ã‚ ²-nitrophenyl-ÃŽ ²-D-glucopyranoside as the substrate according to a previously described method [14]. Endoglucanase and cellobiohydrolase activities were determined by hydrolysis of carboxymethyl cellulose (CMC) and phosphoric acid swollen cellulose (PASC), respectively. PASC was prepared from Avicel PH-101 (Fluka Chemie GmbH, Buchs, Switzerland) as amorphous cellulose. The cell pellet was resuspended in a reaction mixture of 1% CMC or 1% PASC in 50 mM sodium acetate buffer (pH 5.0) with the optical density at 600 nm adjusted to 1.0. After a reaction at 50 ºC for 30 min, the activities were determined by DNS method [15]. One unit of enzyme activity was defined as the amount of enzyme released 1 ÃŽ ¼mol reducing sugar from the substrate per minute. The abilities of the engineered yeast consortium (Y5/EGII + Y5/CBHII + Y5/BGLI) to fermentation ethanol from PASC and steam-exploded corn stover were investigated. The steam-exploded corn stover used in this study was provided by Henan Tian Guan Group Co., Ltd (Nanyang, Henan, China). The raw material was chopped to 2-3 cm size and treated in a steam-exploded vessel at 2.0 MPa for 5 min. The pretreated feedstock was dried at room temperature and directly used as a substrate without washing. The moisture content of the substrate was 8%. The composition of materials was quantitatively analyzed following the NREL Laboratory Analytical Procedure NREL/TP-510-42618 (Structural carbohydrates and lignin) (Sluiter et al., 2008)[16], as shown in Table 3. An enzyme mixture composed of equal amounts of cellulase (Sigma-Aldrich, St. Louis, MO) and ÃŽ ²-glucosidase (Sigma-Aldrich) was used. Yeast cells harboring different surface-display plasmid for EGII, CBHII, or BGLI, were grown in YPD medium a nd then transferred to YPG medium for 48 h at 20 ºC to express cellulase. Cells collected by centrifugation at 5000 rpm for 5 min at 4 ºC, washed with distilled water twice, and mixed in the adjustable ratio to a total initial cell concentration of 30 g/l wet weight to form the functional consortium. Ethanol fermentation proceeded at 30 ºC with 90 rpm in 250 ml Erlenmeyer flasks. 1ml samples of the fermentation broth were taken periodically and stored at -4 ºC until they were analyzed for sugar and ethanol content. The total sugar was determined by the phenol-sulfuric acid method [17]. Glucose was measured by HPLC (model 1260, Agilent Technologies) equipped with a Hi-Plex H column 300 mm Ãâ€" 7.7 mm) and a refractory index (RI) detector. Samples were run at a temperature of 60 ºC and a mobile phase of 5 mM sulfuric acid at a flow rate of 0.6 ml/min. Ethanol analysis was carried out using GC (model 7890A, Agilent Technologies) equipped with a flame ionization detector and a HJ-PEG column. Samples were run under the following conditions: column oven at 120 ºC, front injection port at 200 ºC, with N2 as the carrier gas at a flow rate of 4 ml/min. The expression plasmids pEGII and pCBHII (Fig. 1) were transformed into the yeast S.cerevisiae Y5 strains, respectively. All of recombinant yeast strains had a pAGA1 plasmid for integrating AGA1 into the chromosome, and the resultant transformants were designated strains Y5/EGII and Y5/CBHII (Table S1). Upon galactose induction, the proteins were expected to be secreted and interact with the Aga1p and Aga2p anchor system by using the glycosylphosphatidylinositol (GPI) anchor linked to the cell surface. To confirm displaying of EGII and CBHII on the yeast cell surface, immunofluorescence labeling of the cells was carried out using mouse anti-Xpress IgG antibody as the primary antibody. The green fluorescence of Fuorescein (FITC)-conjugated goat anti-mouse IgG was observed for strains Y5/EGII and Y5/CBHII (Fig. 2), indicating that EGII and CBHII were displayed on the cell surface, respectively. The cells harboring the control plasmids were hardly labeled with mouse anti-Xpress IgG(Fig. 2). These results suggested that two types of cellulase were successfully expressed on the cell surface of S. cerevisiae Y5 strain. As shown in Table 1, EGII, CBHII and BGLI activities were detected in the pellet fraction of strain Y5/EGII, Y5/CBHII and Y5/BGLI, respectively. The strain Y5/CBHII and strain Y5/EGII showed moderate CBHII and EGII activity (1.14 U/OD600 and 1.27 U/OD600, respectively). The BGLI activity of strain Y5/BGLI cells was relatively low, which was only 0.72 U/OD600. No enzyme activity was detected in the culture supernatant (data not shown), and the control strain without displayed enzymes exhibited less than 0.1 U/OD600 of enzyme activity. These results clearly indicated that active enzymes were displayed on the cell surface without leakage into the culture medium. Ethanol fermentation from 10 g amorphous cellulose per liter was performed using a cell combination system consisted of three cellulase-displaying yeast populations. Cells displaying EGII, CBHII and BGLI were mixed in various ratios and the produced ethanol from PASC were measured. S.cerevisiae Y5 without displayed enzymes was the control strain. A mixture of cells with EGII: CBHII: BGLI ratio of 2:1:1 produced the highest amount of ethanol (1.76 g/l) after 84 h; the yield (in grams of ethanol produced per gram of consumed reducing sugar) was 0.42 g/g (Fig. 3). A mixture of cells composed of an equal amount of each cell type produced 0.68 g/l ethanol after 84 h (Figure 3), indicating about 1.6-fold improvement of ethanol production by optimizing the cell ratio. However, a large portion of the substrate (the amount of residual sugar after 84 h hydrolysis of 10 g/l PASC was 5.5 g/l, and the sugar consumption rate was 43.3%) remained after 96 h without being hydrolyzed because the cellu lase activities displaying on cell surface were not enough for complete cellulose digestion. Simultaneous saccharification and fermentation of steam-exploded corn stover (CS) as a sole carbon source was conducted for the cellulase-displaying yeast consortium of the optimized ratio 2:1:1 in the presence of commercial cellulase (Sigma-Aldrich, St. Louis, MO) with different enzyme loadings (0, 0.3, 0.6, 0.9, 1.2, 1.5 FPU/ml). A mixture of cells was incubated in 100 ml of YP medium (20 g/l peptone, 10 g/l yeast extract) for 1 h to remove residual carbon source, and then resuspended in YP-CS medium (YP medium containing 100 g/l steam-exploded corn stover, corresponding to 48.4g cellulose per liter). As shown in Fig. 4, in the presence of 0, 0.3, 0.6, 0.9, 1.2 and 1.5 FPU/ml cellulase, 34.49, 18.71, 7.03, 2.11, 1.98, and 1.23 g/l of residual cellulose remained after 84h, respectively. Addition of 0.9 FPU/ml cellulase enabled utilization of 92.3% of the initial cellulose (Figure 4). The cellulose hydrolyzed by cellulase-displaying yeast consortium with an additional 0.9 FPU/ml cellulase was nearly the same as that by control strain S.cerevisiae Y5 with an additional 1.5 FPU/ml. These results indicate that cellulases displayed on the yeast cell surface improve hydrolysis of cellulose, although their activities were lower than commercial enzymes. Furthermore, using the optimized cell combination system, the relationship between the amount of added cellulase and final ethanol concentration was investigated. As shown in Fig. 5, in the presence of 0.9 FPU/ml cellulase, the cellulase-displaying consortium produced 20.4 g/l ethanol after 72 h, which was similar to the value (20.9 g/l) obtained by control strain in the presence of 1.5 FPU/ml cellulase (Table 2). Notably, as the ethanol yield reached 86% of the theoretical yield with 0.9 FPU/ml cellulase, the cell-surface engineered system enabled a reduction in the amount of added commercial cellulase. Hydrolysis of crystalline cellulose to glucose requires the sequential reactions of three groups of cellulases: endoglucanase, cellobiohydrolase, and ÃŽ ²-glucosidase. CBP is a one-step process where all steps occur in a single reactor and a single microorganism or microbial consortium converts pretreated biomass to ethanol with no additional commercial enzymes. The key challenge of CBP lies in choosing the optimal host to directly convert lignocellulosic materials to ethanol. In recent years, several researchers have been engaged in co-displaying multiple cellulases in a single cell for direct conversion of cellulose to ethanol [18-21]. However, the enzyme activity can be limited because of the metabolic burden [22]. Furthermore, it is difficult to control the surface expression level of each enzyme for optimal ethanol fermentation. Apiwatanapiwat et al., constructed the engineered yeast strain NBRC-5Es that co-displayed two types of amylolytic enzymes, two types of cellulolytic enz ymes (T. reesei EGII and CBHII), and A. aculeatus BGLI on the cell surface. The NBRC-5Es strain produced 1.04 g/l ethanol from 8.44 g/l of the acid-treated Avicel after 48 h of fermentation and resulted in a large portion of the substrate remaining without being hydrolyzed by the enzymes. In this study, instead of co-displaying all the enzymes in one cell, we developed a cellulase-displaying yeast consortium consisting of three types of yeast cells, each displaying different cellulases. This method allows for convenient optimization of ethanol production by adjusting the combination ratio of each cell type for inducing a synergy in cellulose hydrolysis. Diploidization is also a promising strategy for enhancing the fermentation ability of S. cerevisiae. Because polyploid yeast strains, including diploid strains, have higher cell growth rates, cell yields, and tolerances to various stresses compared with haploid strains, they are particularly suited for industrial application. Therefore, to generate an efficient â€Å"whole-cell biocatalyst† yeast strain related to cellulosic ethanol production, we selected S. cerevisiae Y5, a robust diploid strain, as the host cell based on its fermentation and inhibitor tolerance properties [23-24]. We first explored the possibility of ethanol fermentation from PASC by using the surface-immobilized yeast consortium (Y5/EGII+Y5/CBHII+Y5/BGLI). A mixture of cells at the optimized EGII: CBHII: BGLI ratio of 2:1:1 produced 1.6-fold more ethanol (1.76 g/l) than cells composed of an equal amount of each cell type.
